چکیده
The centrosome serves as the primary micro-tubule-organizing center in animal cells and, consequently, functions in many processes, such as migration and formation of the mitotic spindles. The centrosome consists of a pair of centrioles surrounded by pericentriolar material (PCM), the platform for microtubule nucle-ation. The pair of centrioles duplicate once per cell cycle to ensure the equal segregation of chromosomes in mitosis. The control of centro-some duplication and their capacity to nucle-ate microtubules is tightly coupled to cell cycle progression. Centriole duplication initiates at the beginning of the S phase and the duplicated centrioles elongate until the G 2 phase. At late G 2 phase, the centrosomes mature by recruiting PCM components, resulting in the increase in the microtubule-nucleating capacity that helps the formation of spindle microtubules later in mitosis. PCM recruitment in the centrosome maturation process has been intensively investigated and revealed to be regulated by mitotic kinases. However, the mechanism regulating interphase PCM recruitment remains largely unknown, especially in mammalian cells. in other organisms, centriole duplication factors, C. elegans ZyG-1 1 (Plk4 orthlog) and Drosophila Sas-4 2 (CPAP ortho-log) were demonstrated to be involved in the interphase PCM recruitment. in a recent issue of Cell Cycle, Jeffery et al. 3 proposed that centrosomal protein Centrobin regulates microtubule nucleation and organization by controlling the amount of PCM in interphase. Centrobin was initially identified as a daughter centriole-associated protein required for centriole duplication. 4 Centrobin has been shown to have microtubule-bundling activity 5 and plays a role in the stabilization of mitotic spindles by anchoring them to the centrosome, 6 while the role of Centrobin in interphase cells has not been well-defined. First, Jeffery et al. showed that Centrobin is exclusively localized at centrosomes in inter-phase cells 3 in contrast to its association with spindle microtubules during mitosis. They next showed that when Centrobin is depleted in interphase cells, the microtubules become more focused around the centrosome and sparse in the cell cortex area. Furthermore, microtubules are less stable than those in control cells, as detected by sensitivity to microtubule depolymerizing conditions and by the acetylation state of the microtubules. They further demonstrated that altered micro-tubule organization is caused by increase in the number of short microtubules emanating from the centrosome without changes in the microtubule dynamics. Microtubule nucle-ation depends on the amount and integrity of PCM proteins, and Jeffery et al. observed an increase in the intensity of PCM …

An Immunohistochemical Study of Cyclin D1 Expression in Astrocytic Tumors and its Correlation with Tumor Grade
Background & Objective: Glioblastoma-multiforme is the high grade form of astrocytic tumors with a short survival time, which are the most common type of brain tumors. Therefore, finding new therapeutic options is essential. Cyclin D1 is expressed in some human malignancies and can be a potential target for therapeutic intervention. The aim of the present study was to determine...
متن کاملCyclin D1-dependent kinase activity in murine development and mammary tumorigenesis.
Cyclin D1 is a multifunctional protein that activates CDK4 and CDK6, titrates Cip/Kip CDK inhibitors to increase CDK2 activity, and modulates the function of certain transcription factors. To specifically test the importance of cyclin D1-associated kinase activity, we generated "knockin" mice expressing mutant cyclin D1 deficient in activating CDK4/6. The development of several cyclin D1-depend...
متن کاملP-96: Appositional Expressions of Cyclin D1 and E2F1 Gene Machineries in Mycooestrogen Zeralenone-Induced Apoptosis in Testicular Tissue of Rats
Background: Zearalenone (ZEA) is known as a nonsteroidal oestrogenic mycotoxin produced by different species of Fusarium fungi. ZEA is known for its competitive effects with the natural 17-β estradiol to bind with the specific binding sites of the estrogen receptors (Ers). On the other hand, the cyclin family (especially cyclin D1) and E2F1 genes are the checkpoint genes involved in cell cycle....
متن کاملAn In vitro Study on Curcumin Delivery by Nano-Micelles for Esophageal Squamous Cell Carcinoma (KYSE-30)
Background: The incidence of esophageal squamous cell carcinoma (ESCC) is increasing, causing catastrophic health burdens on communities. Curcumin has shown promise as a therapeutic agent in the treatment of colon, colorectal, pancreatic, and esophageal cancers but it has very poor bioavailability. The application of nano-carriers as drug delivery systems increases curcumin's bioavailability. C...
متن کاملp53-mediated growth suppression in response to Nutlin-3 in cyclin D1 transformed cells occurs independently of p21.
Interaction of cyclin D1 with cyclin-dependent kinases (CDK) results in the hyperphosphorylation of the RB family of proteins, thereby inactivating the tumor-suppressive function of RB. Our previous findings suggest that constitutive cyclin D1/CDK activity inhibits p53-mediated gene repression by preventing the appropriate regulation of CDK activity by the CDK inhibitor p21, a transcriptional t...

Background and Aim: The cell cycle is an important event in tumor growth and differentiation and several molecules are involved in this process. The aim of this study was to evaluate the expression of cyclin D1 (a cell cycle inducer) and p21 (a cell cycle inhibitor) in ameloblastoma of the jaws. Materials and Methods: In this cross-sectional study, 40 cases of ameloblastoma were selected from t...
